Clinical summary

Summary

This study is divided into 2 cohorts of people with primary uveal (ocular) melanoma requiring either internal radiation therapy (plaque brachytherapy) or the removal of the eyeball (enucleation). Within each cohort, participants will randomly assigned (by chance) to receive one of two treatments.

In Cohort 1, participants will either receive neoadjuvant treatment with darovasertib followed by brachytherapy, or immediate brachytherapy.

In Cohort 2, participants will either receive neoadjuvant darovasertib followed by definitive primary local therapy (i.e. plaque brachytherapy, proton beam radiation, or enucleation), or immediate enucleation.

Participants will then receive primary local therapy following neoadjuvant darovasertib.

All participants will be followed for up to 3 years to assess longer term outcomes such as vision and tumour recurrence. 

Conditions

This trial is treating people with uveal melanoma who require treatment with either plaque brachytherapy or the surgical removal of the eyeball

Eligibility

Inclusion

  • Primary non-metastatic uveal melanoma
  • Able and willing to provide written, informed consent before initiation of any study-related procedures, and in the opinion of the Investigator, to comply with all study requirements
  • ECOG 0 or 1
  • Adequate organ function

Exclusion

  • Previous treatment for UM
  • Evidence of metastatic UM
  • Attributes that necessitate enucleation regardless of response to therapy
  • Evidence of progressive secondary underlying ocular disease that would confound longitudinal VA assessments
  • Presence of a malignant disease other than the one being treated in this study

Inclusion

  • Your cancer has not spread to other parts of the body (localised).
  • You have been diagnosed with cancer, but have not received any treatment.

Exclusion

  • You have been diagnosed with a prior or secondary type of cancer.
  • You have certain types of non-cancer medical conditions.
  • You have had certain treatments, surgical procedures or drugs.
