Summary
This study is for children, teenagers and young adults aged 25 years and under who have a low-grade glioma (LGG) that has come back or is getting worse.
To take part, the tumour must have a specific gene change (called an alteration) in BRAF or CRAF. These include common changes such as BRAF V600 mutation or KIAA1549:BRAF fusion. These changes are identified through standard genetic testing in certified laboratories.
What treatment is being tested?
All participants receive two cancer treatments:
- Vinblastine (chemotherapy given through a drip)
- Tovorafenib (a targeted therapy taken by mouth)
Both medicines are already used in cancer care. This study is testing the safest and most effective dose when they are used together, and whether the combination can help shrink or control the tumour.
The study has two phases.
Phase A (Feasibility Phase)
This first part focuses on finding the safest and most effective dose of the two medicines when given together.
Participants will receive vinblastine and tovorafenib once a week (on Days 1, 8, 15 and 22). Each treatment cycle lasts 28 days. Participants will have 17 cycles of the combination, followed by 8 cycles of tovorafenib alone.
The dose of tovorafenib may be adjusted (increased or decreased) during this phase based on how participants tolerate the treatment. The research team will check participants' tumours with scans every 3 cycles (about every 3 months).Treatment continues as long as the tumour is not growing and side effects remain manageable.
Phase B (Efficacy Phase)
Once the best dose is confirmed, the study continues using that dose to see how well the treatment works.
Participants receive the same weekly schedule as in Phase A, using the approved dose. The treatment lasts for up to 24 cycles total, just like in Phase A. Scans are done every 3 cycles to monitor how well the tumour is responding. If the tumour grows, side effects become too strong, or you choose to withdraw, treatment will stop.
