Clinical summary

Summary

Eligible participants will be randomly allocated to one of two treatment arms.

In the Experimental Arm, participants will receive sotorasib targeted therapy daily as an oral tablet, plus panitumumab targeted therapy and FOLFIRI chemotherapy regimen every two weeks, both given intravenously (IV)

In the Active Comparator Arm, participants will receive FOLFIRI chemotherapy regimen with or without bevacizumab-awwb, both given via IV every two weeks.

This trial is treating people with KRAS p.G12C mutated metastatic colorectal cancer who have not had prior treatment

Eligibility

Inclusion

  • Pathologically documented metastatic colorectal adenocarcinoma with KRAS p.G12C mutation by a locally validated assay.
  • Central confirmation of KRAS p.G12C mutation
  • Measurable metastatic disease per RECIST v1.1 criteria.
  • Eastern Cooperative Oncology Group (ECOG) Performance Status of ≤ 1.
  • Adequate organ function.

Exclusion

  • Active, untreated brain metastases.
  • Leptomeningeal disease
  • Previous treatment with a KRAS p.G12C inhibitor
  • History of interstitial pneumonitis or pulmonary fibrosis or evidence of interstitial pneumonitis or pulmonary fibrosis on baseline CT scan

Inclusion

  • Your cancer has spread to other parts of the body (metastatic) or has grown into nearby parts of the body (locally advanced).
  • You have been diagnosed with cancer, but have not received any treatment.

Exclusion

  • You have certain types of non-cancer medical conditions.
  • You have had certain treatments, surgical procedures or drugs.
