Clinical summary
Summary
This is a randomised, open-label study with an Experimental arm and an Active comparator arm. In the Experimental Arm, participants will receive Lorlatinib, administered orally once daily, at a dose of 100mg. In the Active Comparator Arm, participants will receive Crizotinib, administered orally twice daily, at a dose of 250mg.

This trial is treating people with ALK-positive non-small cell lung cancer
Eligibility
Inclusion
- Histologically or cytologically confirmed diagnosis of locally advanced or metastatic ALK-positive NSCLC; at least 1 extracranial measurable target lesion not previously irradiated. CNS metastases allowed if asymptomatic and not currently requiring corticosteroid treatment.
- Availability of an archival FFPE tissue specimen.
- No prior systemic NSCLC treatment.
- ECOG PS 0, 1, or 2.
- Age ≥18 years .
- Adequate Bone Marrow, Liver, Renal, Pancreatic Function
- Negative pregnancy test for females of childbearing potential
Inclusion
- You are able to swallow medication by mouth.
- Your cancer has not spread to other parts of the body (localised).
- Your cancer has spread to other parts of the body (metastatic) or has grown into nearby parts of the body (locally advanced).
Exclusion
- You have certain types of non-cancer medical conditions.
- You have had certain treatments, surgical procedures or drugs.
